myBaits version 5 Manual

Basic Procedure:

  1. Prepare library from Illumina kit

  2. Create library pool (7ul template limit per rxn, might need to be done per sample)

  3. Denature library

  4. Hybridize to baits RNA targets

  5. Bind hybridized molecules to magnetic beads

  6. Wash away non-bound library

  7. Elute bound library

  8. Amplify eluted library

Questions:

  1. Are John Calder’s kits available?

  2. How many samples? 200 sample multiplex level, smaller proof of concept

    1. Which Illumina Kit is best? (TruSeq or Nextera). Nextera allows for higher multiplexing, I believe.
