Plate and File Naming Conventions for Library Prep
For ease of processing and transfer of plate names between steps, plates are assigned a systematic plate name consisting of the library number followed by the researchers first and last initials and a number:
4PA1 is in the fourth 16S/ITS library prep, the researcher is Paul A, and it is the first plate created for this run.
Only one plate should be saved per HTX plate reader file; hit “end of batch” after reading absorbances for the second half of the plate. The quantification of the template plus controls should be saved on the petalibrary under MicroLab/DnaQuantification/NovaSeq4/. The above mentioned plate would be 4PA1.xlsx
The nimbus worksheet created for this plate from the above file should have “_DnaConc4Nimbus” (previously “_Whsht”) appended to the name: 4PA1_DnaConc4Nimbus.csv
The normalized plates coming off the Nimbus have “_Norm” appended to the plate name: 4PA1_Norm
PCR1 plates have “_PCR1_MIDplateCode” appended to their names:
4PA1_PCR1_16S0A1, 4PA1_PCR1_16S0B1, 4PA1_PCR1_ITS0A1, and 4PA1_PCR1_ITS0B1
When these plates are combined for the first cleanup, the two MIDplateCodes are appended and the PCR2 plate continues with the same name except PCR2 is used instead of PCR1.
Cleaned Plates Being Stored: 4PA1_PCR1_16S0A1_16S0B1 and 4PA1_PCR1_ITS0A1_ITS0B1
Plates proceeding with PCR2: 4PA1_PCR2_16S0A1_16S0B1 and 4PA1_PCR2_ITS0A1_ITS0B1
